RESUMO

The manufacturing rate of nanoparticles (10-100 nm) is steadily increasing due to their extensive applications in the fabrication of nanoproducts related to pharmaceuticals, cosmetics, medical devices, paints and pigments, energy storage etc. An increase in research related to nanotechnology is also a cause for the production and disposal of nanomaterials at the lab scale. As a result, contamination of environmental matrices with nanoparticles becomes inevitable, and the understanding of the risk of nanoecotoxicology is getting larger attention. In this context, focusing on the environmental hazards is essential. Hence, this manuscript aims to review the toxic effects of nanoparticles on soil, water, aquatic, and terrestrial organisms. The effects of toxicity on vertebrates, invertebrates, and plants and the source of exposure, environmental and biological dynamics, and the adverse effects of some nanoparticles are discussed.

The rapid increase in industrial revolution and the consequent environmental contamination demands continuous monitoring and sensitive detection of the pollutants. Nanomaterial-based sensing system has proved to be proficient in sensing environmental pollutants. The development of novel ligands for enhancing the sensing efficiency of nanomaterials has always been a challenge. However, the amendment of nanostructure with molecular ligand increases the sensitivity, selectivity, and analytical performance of the resulting novel sensing platform. Organic ligands are capable of increasing the adsorption efficacy, optical properties, and electrochemical properties of nanomaterials by reducing or splitting of band gap. Curcumin (diferuloylmethane) is a natural organic ligand that exhibits inherent fluorescence and electrocatalytic property. Due to keto-enol tautomerism, it is capable of giving sensitive signals such as fluorescence, luminescence, ultraviolet absorption shifts, and electrochemical data. Curcumin probes were also reported to give enhanced meterological performances, such as low detection limit, repeatability, reproducibility, high selectivity, and high storage stability when used with nanosystem. Therefore, research on curcumin-modified nanomaterials in the detection of environmental pollution needs a special focus for prototype and product development to enable practical use. Hence, this article reviews the role of curcumin as a natural fluorophore in optical and electrochemical sensing of environmentally significant pollutants. This review clearly shows that curcumin is an ideal candidate for developing and validating nanomaterials-based sensors for the detection of environmental pollutants such as arsenic, lead, mercury, boron, cyanide, fluoride, nitrophenol, trinitrotoluene, and picric acid and toxic gases such as ammonia and hydrogen chloride. This review will afford references for future studies and enable researchers to translate the lab concepts into industrial products.

Ocimum tenuiflorum has been widely used in traditional medicine and has high medicinal value. High volume trade of this potential medicinal plant species led to unscrupulous adulteration of both crude drugs as well as formulations. Morphology-based authentication is difficult in cases of incomplete or damaged samples and in dried herbal materials. In such cases, PCR-based molecular methods may aid in accurate identification. The present study aimed at developing species-specific DNA marker(s) for the authentication of O. tenuiflorum. A species-specific amplicon (279 bp) generated through an inter-simple sequence repeat marker (UBC 835) in all individuals of O. tenuiflorum was cloned, sequenced, and a primer pair was developed (designated as CIM-OT-835F/CIM-OT-835R). The newly developed sequence characterized amplified region marker was validated through PCR amplification in all available seven species of Ocimum, and its specificity for O. tenuiflorum was confirmed with the consistent generation of an amplicon of 177 bp. The developed marker can be used for accurate and rapid identification of the species for certification purposes and will be useful in quality control of medicinal preparations containing this important medicinal species.

The modulatory effect of taurine on 7,12-dimethylbenz(a)anthracene (DMBA)-induced breast cancer in rats was studied. DMBA (25 mg/kg body weight) was administered to induce breast cancer in rats. Protein carbonyl levels, activities of membrane bound enzymes (Na(+) /K(+) ATPase, Ca(2+) ATPase, and Mg(2+) ATPase), phase I drug metabolizing enzymes (cytochrome P450, cytochrome b5, NADPH cytochrome c reductase), phase II drug metabolizing enzymes (glutathione-S-transferase and UDP-glucuronyl transferase), glycoprotein levels, and proliferative cell nuclear antigen (PCNA) were studied. DMBA-induced breast tumor bearing rats showed abnormal alterations in the levels of protein carbonyls, activities of membrane bound enzymes, drug metabolizing enzymes, glycoprotein levels, and PCNA protein expression levels. Taurine treatment (100 mg/kg body weight) appreciably counteracted all the above changes induced by DMBA. Histological examination of breast tissue further supported our biochemical findings. The results of the present study clearly demonstrated the chemotherapeutic effect of taurine in DMBA-induced breast cancer.

High level of morphological as well as chemical variability exists within the genus Ocimum, and its taxonomy and phylogenetic relationships are still doubtful. For evaluating interspecific genetic relationships among the Ocimum species, genotyping with intersimple sequence repeat (ISSR) markers and sequence analyses of noncoding psbA-trnH intergenic region belonging to chloroplast DNA were carried out. Although ISSR markers are highly efficient and reproducible, they have not been used extensively in phylogenetic studies. The use of the plastidial barcode candidate was expected to provide more variable and informative insight into evolutionary rates, and was thus employed as a phylogenetic marker to assess interspecific relationships. This study revealed that the ISSR markers were more efficient than psbA-trnH sequences in resolving the current status of Ocimum L. genus. Distance- and character-based methodological approaches applied on the molecular data with biparental and maternal inheritance were used for deducing the phylogenetic relationships among Ocimum species. Average polymorphic information content (0.344) and resolving power (6.285) depicted through ISSR markers proved to be efficient in discriminating the studied species of Ocimum. The primers used in this study revealed 99.585% polymorphism across the species demonstrating the polymorphic nature of ISSR markers.

Breast cancer is the leading cause of death among women worldwide. Chemoprevention and chemotherapy play beneficial roles in reducing the incidence and mortality of cancer. Epidemiological and experimental studies showed that naturally-occurring antioxidants present in the diet may act as anticancer agents. Identifying the abnormalities of cellular energy metabolism facilitates early detection and management of breast cancer. The present study evaluated the effect of tangeretin on cellular metabolic energy fluxes in 7,12-dimethylbenz(a) anthracene (DMBA)-induced proliferative breast cancer. The results showed that the activities of glycolytic enzymes significantly increased in mammary tissues of DMBA-induced breast cancer bearing rats. The gluconeogenic tricarboxylic acid (TCA) cycle and respiratory chain enzyme activities significantly decreased in breast cancer-bearing rats. In addition, proliferating cell nuclear antigen (PCNA) was highly expressed in breast cancer tissues. However, the activities of glycolytic enzymes were significantly normalized in the tangeretin pre- and post-treated rats and the TCA cycle and respiratory chain enzyme activities were significantly increased in tangeretin treated rats. Furthermore, tangeretin down-regulated PCNA expression on breast cancer-bearing rats. Our study demonstrates that tangeretin specifically regulates cellular metabolic energy fluxes in DMBA-induced breast cancer-bearing rats.

OBJECTIVES: The present study was undertaken to determine the modulatory effect of taurine on the liver mitochondrial enzyme system with reference to mitochondrial lipid peroxidation (LPO), antioxidants, major tricarboxylic acid cycle enzymes, and electron transport chain enzymes during 7,12-dimethyl benz[a]anthracene (DMBA) induced breast cancer in Sprague-Dawley rats. METHODS: Animals in which breast cancer had been induced by using DMBA (25 mg/kg body weight) showed an increase in mitochondrial LPO together with decreases in enzymic antioxidants (superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), glutathione reductase (GR) and glutathione-S-transferase (GST)), non-enzymic antioxidants (reduced glutathione (GSH), vitamin C, and vitamin E), in citric acid cycle enzymes (isocitrate dehydrogenase (ICDH), alpha ketoglutarate dehydrogenase (alpha KDH), succinate dehydrogenase (SDH) and malate dehydrogenase (MDH)), and in electron transport chain (ETC) complexes. RESULTS: Taurine (100 mg/kg body weight) treatment decreased liver mitochondrial LPO and augmented the activities/levels of enzymic, and non-enzymic antioxidants, tricarboxylic acid cycle enzymes and ETC complexes. CONCLUSION: The results of our present study demonstrated the chemotherapeutic efficacy of taurine treatment for DMBA-induced breast carcinomas.

PURPOSE: The aim of the present study was to assess the chemopreventive and chemotherapeutic efficacy of tangeretin on DMBA-induced oxidative stress in breast cancer-bearing Sprague-Dawley rats. METHODS: In this study, the experimental animals were divided into five groups of six animals each. Group I was control, Group II was DMBA-induced breast cancer-bearing rats, Group III was tangeretin pre-treated (50 mg/kg body weight for 30 days orally) breast cancer-bearing animals, Group IV was tangeretin post-treated (50 mg/kg body weight for 30 days orally) and Group V was tangeretin (50 mg/kg body weight) alone treated animals. RESULTS: We have observed the general characteristics of cancer, oxidative stress markers, breast cancer marker, antioxidants and histopathological changes in the experimental animals. We have recorded the body weight, tumor weights, tumor volume and antitumor activity of tangeretin in the experimental animals. Oxidative stress markers, like NO and LPO, and breast cancer marker CEA levels were significantly (p < 0.001, p < 0.05) increased as well as the antioxidants like SOD, CAT, GPx, GST, GSH, ascorbic acid and α-tocopherol were found to be significantly (p < 0.05) decreased in cancer-bearing Group II animals. Whereas, the enzymic and non-enzymic antioxidant levels were found to be significantly decreased in cancer-bearing animals. However, in tangeretin pre-treated and post- treated animals, the levels of antioxidants and breast cancer marker were found to be significantly (p < 0.05) reduced with a concomitant increase in the activities of the antioxidants (p < 0.05). In tangeretin alone treated Group V animals, no significant changes were observed in the levels of antioxidants and breast cancer marker. These results were adequately supported by the histopathological studies in the mammary tissues of the experimental animals. CONCLUSION: From this study, we conclude that the administration of tangeretin was found to be beneficial against DMBA-induced oxidative stress in breast cancer-bearing animals. Hence, we strongly suggest that tangeretin is effective and efficient candidate for the treatment of experimental breast cancer.

Periwinkle (Catharanthus roseus [L.] G. Don), an important medicinal plant, is an allogamous species in which the stigma is below the anthers. The receptive portion is at the base of the stigmatic head and thus automatic intra-flower self-pollination is excluded. The structure of the flower is of typical reverse herkogamy and pollination occurs through nectar-seeking insects. A few self-pollinating strains are also reported in which self-pollination is brought about by an increase in length of the style or of the ovary. Self-pollination is governed by allelic duplicate genes recessive to allogamy. An induced monogenic recessive mutant (EMS 17-1) with caducous closed corolla (corolla abscising before anthesis), isolated from variety, Dhawal, was crossed with two self-pollinating strains to study the possibility of obtaining cleistogamous recombinants combining closed corolla and self-pollination traits. Cleistogamous plants were obtained in which development of fruits and seeds occurred without opening of the corolla. Closed corolla and self-pollination were found to be independently inherited. A dominant gene in the parent in which self-pollination occurred due to an increase in length of the ovary, appeared to completely or partially inhibit expression of the gene for closed corolla in homozygous or heterozygous condition, respectively. The genetic basis of development of cleistogamy is described. Cleistogamy in periwinkle would facilitate in ensuring genetic purity, pollen containment, and seed production even in the absence of pollinators. This appears to be the first report on the development of cleistogamous plants in an allogamous species.
